abstract="An analysis was made of erythroid colony-forming unit(CFU-E) and erythroid burst-forming unit(BFU-E) in bone marrow of LACA mice treated with harringtonine (Ha) using a methyl cellulose gel culture system. Increased numbers of CFU-E and BFU-E were observed 24 hours after treatment with a single dose of Ha at 0.5, 1.0 and 1.5mg/kg. Then the numbers decreased while dose of Ha increased. But a stimulatory effect on the proliferation of CFU-E and BFU-E still was found after 3,6,9,12 and 15 consecutive daily treatment with 1.0mg/kg of Ha. The conditioned medium from bone marrow of Ha-treated mice had no effect on the numbers of CFU-E and BFU-E. The time-response curve showed there were two peaks in numbers of CFU-E and BFU-E on days 1 and 12 after treatment. Analysis of proliferation by the cytosine arabinosis suicide technique indicated a significant increase in the percentage of CFU-E or BFU-E in S-phase at 12 and 24 hours after treatment.
