N2  - The examination of fingernails and toenails is increasingly becoming the focus of research as an alternative for the detection of chronic or past intake of substances. The development and validation of an LC-MS/MS method for the detection of the alcohol consumption marker ethyl glucuronide (EtG) in nails and hair is presented. This was used in a comparative study of EtG storage in hair and nails with samples from a group of living normal drinkers (n= 18) and post-mortem samples from a group of fatalities (n= 19) with known chronic excessive alcohol consumption. For the group of living subjects, an average higher EtG concentration was found in nails than in hair. In the group of deaths there were larger fluctuations in the EtG findings, which can possibly be explained by influences caused by the position (lying in body or putrefactive fluid). For proof of absolute abstinence, however, nails are a sensible alternative.   ===  Die Untersuchung von Finger- und Fussnaegeln rueckt zunehmend als Alternative fuer den Nachweis der chronischen beziehungsweise zurueckliegenden Aufnahme von Substanzen in den Fokus der Forschung. Vorgestellt wird die Entwicklung und Validierung einer LC-MS/MS-Methode zum Nachweis des Alkoholkonsummarkers Ethylglucuronid (EtG) in Naegeln und Haaren. Diese wurde im Rahmen einer vergleichenden Untersuchung der Einlagerung von EtG in Haaren und Naegeln mit Proben einer Gruppe lebender Normaltrinker (n= 18) sowie post-mortem Proben von einer Gruppe von Todesfaellen (n= 19) mit bekanntem chronisch-exzessivem Alkoholkonsum eingesetzt. Fuer die Gruppe der lebenden Probanden konnte eine durchschnittlich hoehere EtG-Konzentration in Naegeln im Vergleich zu den Haaren gefunden werden. In der Gruppe der Todesfaelle zeigten sich groessere Schwankungen der EtG-Befunde, die moeglicherweise durch lagerungsbedingte Einfluesse (Liegen in Koerper- oder Faeulnisfluessigkeit) erklaer bar sind. Fuer einen Beleg absoluter Abstinenz sind Naegel jedoch eine sinnvolle Alternative.<p /> <p>Language: de</p>
