Abstract

An ELISA was developed in analogy to the standard practice of immuno- chemical pre-testing of drugs and pharmaceutical substances. It was developed for the detection of ethyl glucuronide (EtG). The test kit's usefulness was examined using native sample materials as a screening test. This required the determination of EtG by means of ELISA using 22 anonymous urine samples as well as 15 serum samples, which were then compared with GC/MS results. In 7 cases deviations of below 30 % could be shown. False positive results could not be found. In 2 cases a false negative result could be found due to the high cut-off value. Measurements by means of ELISA tended to be slightly higher at a cut-off value of 1 and in 2 cases they even showed a false positive result with alcohol free samples. The pilot study presented an accuracy of approximately 71% and a sensitivity of 100 %. Accuracy could be improved by selection of antibodies.
